Learning objectives
Il corso integrato affronta le principali problematiche relative alle biotecnologie applicate ai gameti e alle fasi precoci dello sviluppo embrionale. Gli studi che affrontano gli aspetti molecolari dell'embriologia avanzata, le modalità specifiche che regolano il differenziamento tissutale e la capacità replicativa della cellula pluripotente (sviluppati nel modulo 1), trovano la loro logica prosecuzione e completamento con l'apprendimento delle metodiche di micromanipolazione dei gameti e degli embrioni e di valutazione della qualità del seme per il suo impiego nelle moderne biotecnologie per la cura dell'infertilità.

Key molecules in early embryo development (1 hour)
Trofectoderm and inner cell mass. Different contribution to stem cell derivation (2 hours)
Imprinting and epigenetic regulation (1 hour)
Maternal and paternal contribution to the mitotic spindle (2 hours)
Compartimentaliation of degli organelles (2 hours)
Microinjection and transgenesis (1 hour)
Nuclear transfer (1 hour)
Quantitative gene expression in early embryo (2 hours)
Morfogenetic gradient and RNA sorting in oocytes and embryos (2 hours)
Post-transcriptional control in mammalian oocytes (2 hours).

Practical training
Staining cells for pluripotency genes (2 hours)
Differential staining (3 hours)
Centrioles /centrosome localization(3 hours)
Use of standards in PCR of oocytes and embryos (3 hours)
Staining of cell compartments (3 hours)
Use of micromanipulation and computerized injector (2 hours)
