Obiettivi formativi
The educational objective of the course is to introduce the students to the comprehension of the analytical genomic tools and to the mechanisms and machineries of inherited and parasitic diseases. Some host-parasite molecular mechanisms of interaction will also be considered. The course will also illustrate the spontaneous and experimental animal models for the understanding of the genetic and pathogenetic basis of parasitic diseases.
Risultati apprendimento attesi
At the end of the course the student will possess:
-knowledge on the principles of the construction of the genetic and physical maps
- knowledge of the main tools available for nucleotide sequencing both Sanger and next generation
-knowledge of the main tools for high-throughput genotyping both by hybridization and sequencing
- basic knowledge of the genomic difference in parasites and mammals
Furthermore the student will be able to
- understand the terminology and the research approaches of molecular genetics and genomics used in science and reported in the scientific publications
- understand which tools might be more appropriate for an analysis aimed to high-throughput nucleotide sequencing and genotyping, together with their pros and cons
- understand which tools might be the most appropriate for an association study for different diseases, and which might be the limits and the power.

Unit H52-79-A: Genomic analysis of inherited diseases
LECTURES:
- Genetic and physical maps of domestic animals (2 hs).
- Whole genome reference sequences (1h)
- Nucleotide sequencing: Sanger and High throughput Sequencing methods (3 hs)
- Genome-wide SNP detection (for microarray) (1h)
- Genotyping by Sequencing (1h)
- Genotyping by hybridization (microarray) and by whole genome sequencing (1h)
- Linkage/Association analyses using genomic SNP genotypes: family (HM) and cases/controls studies (GWAS) (2h)
- Genomic variability and host-parasite interaction (1 hs)
PRACTICALS:
- Online practice on nucleic acid quantification for genomic analyses. Online practice on Genomic databases (4).
- Visits to research and service centres equipped with high-throughput platforms when available /or computer practice for GWAS analyses (4).
- Journal clubs on genome sequencing projects in domestic animals, gene mapping and identification in inherited diseases and parasitic diseases, the latter limited to those addressed in the lectures (4).

Unit H52-79-B: Molecular pathology of parasitic diseases.
LECTURES:
Unit 1 - Leishmaniasis: immunopathology and primary animal models (4hs)
Unit 2 - Individual and social honey bee immunity against parasites (2hs)
Unit 3 - Moonlighting proteins of parasites (2hs)
Unit 4 - Neospora and Toxoplasma - Pathology and host range determinants (2hs)
Unit 5 - Drug targets of parasites - insights from genome mining (2hs)
PRACTICALS:
Unit 1 - MicroRNA web resources (4hs)
Unit 2 - MicroRNA analysis - RNA extraction and retrotranscription (4hs)
Unit 3 - MicroRNA analysis - Quantitation (4hs)
Prerequisiti
No specific prerequisites are required.
Metodi didattici
Lectures and practical sessions (the latter consist of seminars, visits to external labs, and computer activities)

Modalità di verifica dell’apprendimento e criteri di valutazione
Written examination: The test consists of open-ended texts and multiple-choice questions concerning the topics of both Units. The student must pass both Units on the same day. The exam will be held in Lodi, in a computer lab using Microsoft Forms.
The evaluation will take into account the quality and the accuracy of the student's open-ended text answers, including the adequate motivation of statements and analyses.
