Learning objectives
Acquisire strumenti e competenze sulla possibilità di utilizzare strategie a basso impatto ambientale
(Microorganismi antagonisti (BCA) o molecole di dsRNA) per il controllo di patogeni e parassiti delle piante.
Subunità 1: Preparazione strumenti di controllo a basso impatto ambientale
a) Isolamento e caratterizzazione di BCA nei confronti di differenti agenti patogeni.
b) Quantificazione del BCA/patogeno da inoculare nelle prove successive in serra mediante digital PCRo

c) Costruzione di molecole di dsRNA per il controllo di virus (TAV e virus della vite) e funghi (es. Botrytis
cinerea )
d) Quantificazione di dsRNA mediante digital PCR
Subunità 2: Prove in ambiente controllato e rilievi
fenotipici
a) Trattamenti con BCA e dsRNA su piante inoculate sperimentalmente con i patogeni sopra descritti
b) Rilievi fenotipici; analisi visive, determinazione quantità clorofilla a mediante fluorimetro
c) Analisi statistiche
Subunità 3: Determinazione dell'effetto sulla pianta Le piante inoculate con gli agenti patogeni verranno
analizzate con tecniche molecolari
a) Quantificazione dei patogeni inoculati, del BCA e del dsRNA mediante Digital PCR
b) Analisi statistica
Expected learning outcomes
Al termine del corso gli studenti avranno acquisito conoscenze e competenze sulle metodiche di laboratorio per lo studio di patogeni mediante tecniche molecolari e il controllo dei patogeni utilizzando microorganismi antagonisti o molecole dsRNA.

Subunit 1: preparation of low environmental impact control tools
a) isolation and characterization of BCA against different pathogenic agents
b) quantification of BCA/pathogens to be inoculated in the following greenhouse trials by use digital PCR and/or microscopy
c) synthesis of dsRNA molecules for the control of viruses (TAV and grapevine viruses) or fungi (e.g. Botrytis cinerea)
d) quantification of dsRNA by digital PCR

Subunit 2: trials in controlled environment and phenotype observation
a) treatments with BCA and dsRNA on plants experimentally inoculated with the pathogens previously described
b) phenotype observation: visual examination of symptoms, quantification of chlorophyll through fluorimetric analysis
c) statistical analysis of data
Subunit 3: Detection of treatment effect on the plants
a) quantification of pathogens, BCA, and dsRNA in inoculated plants by digital PCR
b) statistical analysis of data
