Obiettivi formativi
This course aims to provide an in depth understanding of the fundamental principles guiding molecular genetics approaches in either model organisms or human research. By the end of this course students will be able to critically evaluate practical solutions to complex genetic problems by applying the principles of transmission genetics.

1) Complex genomes and hereditability of complex traits
Genetic and physical maps: DNA polymorphisms as markers.
Positional cloning, from phenotype to gene: Duchenne muscular dystrophy as a case study. Genetic, cytogenetic and physical mapping, cloning and expression analysis of DMD gene and its mutations, clinical applications.
Functional genomics, from sequence to gene function. Arabidopsis as model system for plant biology. Tools for functional genomics. Sensitised genetic background screens. Quantitative effect of mutations, molecular basis of genetic dominance. Genome-enabled genetics and genome wide association mapping.
Research approaches to nutrigenomics, genomic imprinting and examples of nutriepigenetics.

2) The genetics of differentiation
Sex determination in Drosophila as multilevel regulatory system.
Sex determination in mammals. Role of SRY and SOX9. Mutations determining "sex reversal".
Dosage compensation: hyperactivation and inactivation of chromosome X.
Genetic dissection of the signal transduction determining the transition from vegetative to reproductive phase.
In addition, every year a monographic topic will be proposed.

Propedeuticità
According to the rules of the graduate programme in "Molecular Biology of the Cell".
Prerequisiti
An intermediate level of understanding of Mendellian genetics and molecular biology is mandatory.
Evaluation: Final written exam including open question and multiple choices tests.
